Genetic analysis of sex-chromosome arrangement in Drosophila americana: a laboratory exercise for undergraduate or advanced placement

ثبت نشده
چکیده

This paper describes a crossing design for determining the arrangement of the X and 4 chromosomes in Drosophila americana. Implementation of this crossing design in a teaching laboratory with undergraduate or advanced placement students provides a clear demonstration of sex-linked vs. autosomal inheritance and the connection between linkage relationship and chromosomal arrangement. Analyses can be performed on newly collected material, or on established stocks available from the Tucson Stock Center (University of Arizona) or the author. The procedure requires minimal setup, takes about 10 weeks to complete, and generates easily interpretable results. In this description of the procedure, an example is provided where four undergraduate students analyzed a sample (OR01) of D. americana collected near Toledo, Ohio. An electronic copy of the procedure is available from the author upon request ([email protected]).

Positive selection near an inversion breakpoint on the neo-X chromosome of Drosophila americana.

Unique features of heteromorphic sex chromosomes are produced as a consequence of sex-linked transmission. Alternative models concerning the evolution of sex chromosomes can be classified in terms of genetic drift or positive selection being the primary mechanism of divergence between this chromosomal pair. This study examines early changes on a newly acquired chromosomal arm of the X in Drosop...

A concise Drosophila laboratory module to introduce the central concepts of genetics

The quick generation time, ease of handling, and wide availability of striking phenotypic mutants makes Drosophila melanogaster a highly attractive system to expose undergraduate and advanced high school students to concepts in genetics. However, it is nevertheless very labor intensive to set up Drosophila for a large scale laboratory exercise in a short period of time. The previously developed...

Chromosomal Analysis of Couples with Repeated Spontaneous Abortions in Northeastern Iran

Background Cytogenetic study of reproductive wastage is an important aspect in determining the genetic background of early embryogenesis. Approximately 15 to 20% of all pregnancies in humans are terminated as recurrent spontaneous abortions (RSAs). The aim of this study was to detect chromosome abnormalities in couples with RSAs and to compare our results with those reported previously. Materia...

The First Case of a Small Supernumerary Marker Chromosome 18 in a Klinefelter Fetus: A Case Report

Small supernumerary marker chromosomes (sSMCs), or markers, are abnormal chromosomal fragments that can be hereditary or de novo. Despite the importance of sSMCs diagnosis, de novo sSMCs are rarely detected during the prenatal diagnosis process. Usually, prenatally diagnosed de novo sSMCs cannot be correlated with a particular phenotype without knowing their chromosomal origin and content; ther...

Reduced sequence variability on the Neo-Y chromosome of Drosophila americana americana.

Sex chromosomes are generally morphologically and functionally distinct, but the evolutionary forces that cause this differentiation are poorly understood. Drosophila americana americana was used in this study to examine one aspect of sex chromosome evolution, the degeneration of nonrecombining Y chromosomes. The primary X chromosome of D. a. americana is fused with a chromosomal element that w...
